Generation of gene targeting medaka(遺伝子ターゲティングメダカの作成)

My work is focusing on conventional gene targeting via ES-mediated homologous recombination approach, as well as ZFN-mediated gene targeting in medaka.

We previous performed conventional gene targeting in medaka diploid and haploid ES cell line (MES1 and HX). By transfection of a targeting vector containing reporter cassette, we successfully disrupted medaka Nanos3 locus via homologous recombination, and isolated mutant cells with HR through positive-negative drug selection. Recently, we use engineerized ZFN to target disrupt gsdf, an essential gene playing roles in sexual differentiation in medaka.

Nanos gene targeting work is now on revised submission, and gsdf gene targeting is under preparation.
