Transformed rice cells were partially synchronized by aphidicolin treatment and then released into the S phase. Accumulation and turnover of the mRNA transcribed from the wheat Hl promoter/GUS fusion gene was analyzed by the S1 protection assays. Ouantity of mRNA was corrected for that of constitutively transcribed HPH(hygromycin phosphotransferase) mRNA. DNA synthesis rates were monitored in parallel by measuring the incorporation of [3H]TdR. A, accumulation and turnover of the transcript from the 5'DELTA-771 (full size) promoter/GUS fusion gene; B, accumulation and turnover of the transcript from the 5'DELTA- 153 H1 promoter/GUS fusion gene.
